【摘要】:Protectionofaquaticspeciesrequirespreventionofunacceptableeffectsonpopulationsinnaturalhabitats.Toxicitytestsareconductedtoprovidedatatopredictwhatchangesinviablenumbersofindividualspeciesmightresultfromsimilarexposureinthenaturalhabit.Informationmightalsobeobtainedontheeffectsofthematerialonthehealthofotherspecies.Bioluminescentdinoflagellatesrepresentanimportanteucaryoticgroupwhicharewidelydistributedintheoceanicenvironment.1.1Thisguidecoverstwodistinctprocedures,basedonsimilarprinciples,forobtainingdataconcerningtheadverseeffectsofatestmaterial(addedtodilutionwater)onoceanicbioluminescentdinoflagellates.1.1.1Theendpointforbothproceduresisbasedonameasurablereductionorinhibitioninlightoutputfromthedinoflagellates.Bothproceduresaresimilarinthatwhenbioluminescentdinoflagellatesareexposedtotoxicants,ameasurablereductioninbioluminescenceisobservedfromtheircellsfollowingmechanicalstimulationwhencomparedtocontrolcells.Inthefirstprocedure,cellsofthebioluminescentdinoflagellateGonyaulaxpolyedracanbetestedoverarangeofuptosevendaysofexposure(orlonger)toatoxicant.Thesecondprocedureusesanotherspecies,Pyrocystislunula,fora4htest.1.2Bothprocedurescanmeasurethetoxiceffectsofmanychemicals,variousmarineandfreshwatereffluents,antifoulingcoatings,leachates,andsedimentstobioluminescentdinoflagellates(1-5).Compoundswithlowwatersolubilitysuchaslargeorganicmoleculesmaybesolubilizedwithmethanol,ethanol,andacetonesolventsfortesting(4)(seeGuideE729).1.3AnIC50inlightoutput(bioluminescence)istherecommendedendpoint(1).However,percentinhibitionofbioluminescenceisanappropriateendpointinsomecases(5).1.4Othermodificationsoftheseproceduresmightbejustifiedbyspecialneedsorcircumstances.Althoughusingappropriateproceduresismoreimportantthanfollowingprescribedprocedures,resultsoftestsconductedusingunusualproceduresarenotlikelytobecomparabletoresultsofothertests.Comparisonofresultsobtainedusingmodifiedandunmodifiedversionsoftheseproceduresmightprovideusefulinformationconcerningnewconceptsandproceduresforconductingacuteandchronictests.1.5ThevaluesstatedinSIunitsaretoberegardedasthestandard.1.6Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
